Again the dilemma of home testing verses testing at your repo vet for your bitches levels.
I have done both. If it is a bitch that I am very familiar with and will be doing an AI I will do a Vaginal Cytology instead of a progesterone test. I have two bitches that are like clock work and a smear has always been just fine for them. I am looking for cornification of the cells to see where they are along with their signs of readiness to be bred.
I however have and still use the home testing kits such as Target and Pre Mate. It is a simple blood draw and you let the serum seperate from the whole blood and follow the directions. This along with a vaginal Cytology has worked like a charm for me with fresh chilled being shipped in.
I do believe that with a young bitch that is not very reliable with their heat cycles and have never been bred that an initial base line should be established. This I have done at my repo vet with a blood draw to be sent to the lab to give me an actual progesterone level. I follow this and will have it sone up to three times in the heat cycle to see how my bitch is climbing and when she has her LH Surge.
It is always good to use your repo vet if you are doing a surgical insemination using frozen semen. That is for another blog
I routinely use vaginal cytology as it is minimally invasive and I know results very quickly *less than five minutes*
